About FOXO4-DRI

FOXO4-DRI (sometimes Proxofim) is a synthetic D-retro-inverso peptide derived from the FOXO4 transcription factor. Its mechanism targets the p53-FOXO4 interaction that keeps senescent cells alive — by disrupting this interaction, FOXO4-DRI selectively induces apoptosis in senescent cells while sparing healthy cells. It’s a senolytic, the first peptide-based compound to show this effect in a published animal-model study (de Keizer lab, 2017).

This is one of the most experimental compounds in the library. Human safety and efficacy data is essentially absent — published research is animal-model only. Anyone working with FOXO4-DRI for research purposes is operating well outside established clinical literature. Treat findings, dosing, and protocols with appropriate caution.

Plasma half-life for FOXO4-DRI is poorly characterised in published literature — animal studies suggest a short plasma window with longer tissue retention typical of D-retro-inverso peptides (resistant to proteolytic degradation). What’s a senolytic?

A compound that selectively kills senescent cells — cells that have stopped dividing but resist apoptosis and contribute to age-related inflammation. The concept is that clearing these cells may reduce age-related pathology. Other senolytics studied in research include dasatinib + quercetin combinations and fisetin.

Is human data available?

Almost none. The published work is animal-model based. Anyone using FOXO4-DRI for human research is essentially in unstudied territory. Conservative dosing and cautious assessment are appropriate.

How is it different from other anti-aging peptides?

Most anti-aging research compounds work by improving cell function (NAD+ pathway, autophagy, telomere maintenance). FOXO4-DRI works by removing cells — specifically senescent cells. The mechanisms are complementary rather than competing.

What injection site is used?

Subcutaneous abdominal is the documented route in extrapolated protocols. Given the experimental nature and limited data, site rotation is sensible but standardised protocols don’t exist.
